Hoffmeister Series Ions Protect Diphtheria Toxoid from Structural Damages at Solvent/Water Interface

چکیده
During the W1/O phase (in the W1/O/W2 process) of protein microencapsulation within poly-lactide-co-glycolide (PLGA), hydrophobic interfaces are expanded where interfacial adsorption occurs followed by protein unfolding and aggregation. Spectroscopic and immunological techniques were used to ascertain the effects of the Hoffmeister series ions on Diphtheria toxoid (Dtxd) stability during the W1/O phase. A correlation was established between salts used in aqueous solutions and the changes in Dtxd solubility and conformation. The Dtxd α-helical content was quite stable thus leading to the conclusion that encapsulation was followed by protein aggregation, with minor exposition of hydrophobic residues and a small change at the S-S dihedral angle. Dtxd aggregation is 95% avoided by the chaotropic SCN. This was used to prepare a stable Dtxd and immunologically recognized/PLGA formulation in the presence of 30 mM SNC. The recovery increased by 10.42% or 23.2% when microencapsulation was within the COOMe or -COOH (12kDa) PLGA, respectively. In conclusion, the aim of this work was achieved, which was to obtain the maximum of Dtxd stability after contact with CH2Cl2 to begin its PLGA microencapsulation within ideal conditions. This was a OPEN ACCESS Materials 2009, 2 766 technological breakthrough because a simple solution like salt addition avoided heterologous proteins usage.

The Effect of Hoffmeister Salts on the Chaperoning Action of β-Casein in Preventing Aggregation of Reduced β-Lactalbumin
Protein aggregation and precipitation is associated with many debilitating diseases including Alzheimer's, Parkinson's, and light-chain amyloidosis. β-Casein, a member of the casein family, has been demonstrated to exhibit chaperone-like activity to protect protein form aggregation. Hofmeister salts (lyotropice series) are a class of ions which have an effect on the solubility and also the stab...
متن کاملDevelopment of a freeze-stable formulation for vaccines containing aluminum salt adjuvants.
Vaccines containing aluminum salt adjuvants are prone to inactivation following exposure to freeze-thaw stress. Many are also prone to inactivation by heat. Thus, for maximum potency, these vaccines must be maintained at temperatures between 2 degrees C and 8 degrees C which requires the use of the cold chain. Nevertheless, the cold chain is not infallible. Vaccines are subject to freezing duri...
متن کاملPreparation and Evaluation of Diphtheria Toxoid-Containing Microspheres
Preparation of chitosan (CS) microspheres as a novel drug delivery vehicle for intranasal immunization using high, medium and low CS molecular weight (MW) was investigated in this study. Diphtheria toxoid (DT) was used as a model antigen. The emulsion-solidification method was adopted for microencapsulation of DT. In the first step, following the purification of semi-crude DT by the ion-exchan...
متن کاملEffectiveness of Vaccination During Pregnancy to Prevent Infant Pertussis.
BACKGROUND Vaccination against pertussis during pregnancy is recommended to protect newborns, yet there is limited information about the effectiveness of maternal tetanus toxoid, reduced diphtheria toxoid, acellular pertussis (Tdap) vaccine before the first infant dose of diphtheria, tetanus and acellular pertussis (DTaP) vaccine and during the first year of life in infants who have received DT...
